Remove Antivirus Pro 2010 – How To Uninstall Antivirus Pro 2010 (Antivirus Pro 2010 Removal Guide)

Do you want to Remove Antivirus Pro 2010? Antivirus Pro 2010 is a misleading security application pushed through Braviax malware (Trojan.Virantix.C Trojan virus).

This program is classified as misleading application because it reports false scan results and displays fake security alerts to make you think your computer is infected. AntivirusPro 2010 is from the same family as PC Antispyware 2010, Home Antivirus 2010 and PC Security 2009.

Once installed, the misleading application will ask you to pay for a full version of the program to remove non-existing system security threats. Do not buy this program. Uninstall Antivirus Pro 2010 from your computer as soon possible.

How To Completely Remove Antivirus Pro 2010?

Typically the standard uninstall measures do not fully remove Antivirus Pro 2010. So, if you have already tried this via the usual Add/Remove Programs via the Control Panel, and it does not work, then the only other thing to do is to remove it manually. You could also use a utility program to this this for you – we’ll discuss this later.

What To Do If The Uninstall Fails, Or You Don’t Have A Clue How To Do It Manually?

Not to worry if you still see signs of Antivirus Pro 2010 on your computer.  There’s a simple answer.  You may still have some other files left over from the installation which now need to be deleted manually. These would normally include windows registry files, associated dlls and folders.

As you can imagine, deleting all the files mentioned above manually would be very time consuming and frustrating, and would probably prove unsuccessful, since most likely you won’t even know the names of the files, and where they are located.  This could be dangerous and prove more harmful than helpful.

The best way to be 100% sure that you will remove Antivirus Pro 2010 from your computer would be to use a utility software that specializes in this process.

The Solution: An Uninstall Program

There are many of these utility programs available online, but you will be wise to try to find one that has some merit and good reviews by experienced computer users, as well as, maybe having had some prestigious awards tacked on to it from the computer software industry.  This is a sure winner.

Consider and ask yourself whether such an uninstall program will do the trick:

•will it completely remove any software installed (in this case, remove Antivirus Pro 2010) ?
•will it be much faster than Add/Remove program ?
•can it uninstall system and hidden programs ?
•does it offer different ways of sorting applications e.g. by name, occupied size, installation date ?
•does it have the capability of uninstalling programs that could not be removed by Add/Remove programs ?
•will it force removal of uninstall related entries in the registry

Why don’t you take a look at a top uninstaller? Visit: Perfect Uninstaller

An uninstaller which is highly recommended and very popular is Perfect Uninstaller – its free to try and has great and rave reviews so you should check it out as a possibility.  What do you have to lose?  Only your nagging problem when you are finally able to remove Antivirus Pro 2010.

So, do you want to remove Antivirus Pro 2010? Today? Right now? Visit: Uninstall Antivirus Pro 2010

IBM Acquisition of Sun Microsystems – Trading JAVA

With the doubling of the JAVA stock price on March 18, 2009 based on a Wall Street Journal report that IBM is bidding to buy Sun Microsystems, and with the StockTradersPlace sell action commentary on March 21, one open question is “Should the stock be shorted?”. Here is where other factors have to be considered beyond technical analysis. With speculation that other companies such as Hewlett-Packard (HPQ) and Cisco Systems (CSCO), among others, may want to buy Sun Microsystems, a bidding war can emerge. For this reason, it is probably risky to short the stock at this point, although selling the long position holding is the prudent thing to do.

I continue to be amused by corporate acquisitions and the price paid for such actions. I was involved in such a scenario in 2001 (at the onset of the tech/telecommunications industry meltdown) where the company I was working for ballooned to 95,000 employees largely on the contribution of various corporate acquisitions leading up to year 2001. Today, that company has less than 32,000 employees and has filed for bankruptcy protection.

The previous paragraph was not meant to suggest a similar fate for IBM but was simply old memories coming to the fore as this Sun Microsystems acquisition is being reported. The price is high at $8B. There will be corporate culture clashes and integration issues, as well as clear conflicts in technology in terms of Intel and Advanced Micro Devices microprocessors used in IBM server products versus the Sun Microsystems SPARC architecture microprocessors.

And what is the latest craze with Sun Microsystems anyway? Well, Sun’s legacy is in engineering workstations and server products. But in 2007, Sun changed its stock symbol from SUNW to JAVA to clearly re-orient the company strength and leadership in the JAVA programming language. As significant as the JAVA technology is to the Web computing industry, one must continue to question how they intend to make money from this. As an investor/trader, that is the most important question to answer.

It will be very interesting indeed to see how this unfolds if IBM does end up acquiring Sun Microsystems. Will IBM then customize JAVA for its own internal purposes, thereby creating a potential issue with the open JAVA standard? How does IBM intend to recover the $8B spent predominantly on the acquisition of the JAVA technology which rightly belongs in the domain of open standards.

As an investor/trader of JAVA now and IBM now and in the future, the company fundamentals are extremely important to analyze beyond the technical analysis indicators. This is the time to avoid being over-focused on technical analysis alone.

Grey’s Anatomy Season 7 Premier

Watch Grey’s Anatomy Season 7 Episode 1. The final episodes of Season 6 found Owen and Cristina juggling their commitment to each other with the unsettling return of painful memories for each of them. While Teddy’s presence had both provided a teacher for Cristina and given Owen a chance to regain a friendship, it had also triggered his wartime memories.

With Cristina’s encouragement, Owen settled into working alongside Teddy in Seattle as they had in the field. Due to this experience, however, we soon began to see Owen again experiencing PTSD symptoms except this time he also flashed back to the painful loss of a comrade in the field. As his memories of that loss unfolded over the course of a powerful episode we saw both the derivation of his friendship with Teddy and gained insight into the likely event Owen was dreaming about when he choked Cristina the previous season during a PTSD-triggered night terror.

Owen became increasingly erratic both at home and at work, causing Cristina to express her concern while also mystifying Teddy who found her friend suddenly unwilling to back up her medical choices as he would have in the field. While this experience was exceedingly painful for all involved, Owen also found some relief through talking to the husband of a patient being treated by Teddy and Cristina. His wife had requested physician assisted suicide as cancer ravaged her frail system. Over the course of an emotionally tenuous day Owen realized the parallels between his stressful response to the patient’s choice and his inability to save his comrade during the war.
(click read more after the image to continue)

As Owen reached this realization he sat with the patient’s husband and said, ‘There will be a moment, when she dies. Her face will relax and all that pain she was in will be gone. And you’ll feel relief, relief for her, relief for you. You will know with absolute certainty that you did the right thing…You have to hold onto that moment because all the rest of it is just your own garbage.’

Both Cristina and Teddy struggled to assist Owen from their very different vantage points. Cristina tried to provide opportunities for Owen to talk while asking him to consider a return to therapy. After Owen bristled at her suggestions Cristina took matters into her own hands by asking for assistance from Teddy, the friend who had been there alongside Owen as events took their toll on him in Iraq. Teddy, aware that in this difficult situation the only way to help Owen was to help Cristina help him, offered Cristina simple advice, ‘He’s in love with you…Show him how you feel.’

Upon reaching home to find Owen attempting to return to some sense of normalcy by observing the basic rituals of making dinner, Cristina felt momentarily hopeful. Yet Owen’s acute reaction to something burning on the stove caught them both off guard. When Owen reached for Cristina she flinched, forcing a painful realization for them both. ‘You’re scared of me,’ he whispered as a broken expression flashed across his face. Cristina admitted, ‘I don’t want to be,’ before reaching for Owen’s hand. And with that, Owen accepted that a return to Dr. Wyatt’s psychiatric couch was imminent.

As he began to allow himself greater access to his wartime memories in order to heal from them Owen found himself facing a conundrum. The past he’d disavowed was intent on coming to the surface and he discovered the value of those memories amidst the pain they generated. The experience of working as a trauma surgeon in Iraq had provided him opportunities for growth and important friendships with Teddy and the other members of their platoon. At the same time that he had attempted to place those memories behind a locked door, they also formed the framework for the kind of man, the kind of surgeon he had become as a result of those experiences and it was that man who had fallen in love with and was loved in return by Cristina. Yet Owen continued to fear how those memories would impact Cristina as he struggled to face the future with her while making peace with the past.

In an interesting turn of events, we soon found Teddy never told surgical department administration she’d stopped looking for a job elsewhere. Having opened a national search for a head of cardio thoracic surgery, Derek squired eminent heart surgeon Tom Evans around the hospital. While Cristina was excited at the opportunity to observe a prominent cardio god in action, Teddy flailed as she began to fight hard for a job that had become important to her. It unfolded across a tense episode that Teddy valued both the new version of her friendship with Owen as well as opportunities to work with Cristina. Yet Owen was saddened at watching Teddy’s miserable struggle to retain a job she’d taken on at his request. Adding additional tension was Cristina’s request that Owen endorse Teddy to Derek in an attempt to sway him to offer Teddy a permanent job.

When Owen discovered Derek had already chosen to offer the contract to Dr. Evans, Owen found himself standing at a crossroads. As Cristina had regained her professional confidence, Owen believed her enthusiasm about Evans’ appearance indicated she was now in a position to learn from any qualified mentor. Owen also seemed to wonder if his best course of action to fix what was broken amidst his friendship with Teddy was to assure Derek she had opportunities elsewhere, potentially giving her a chance at a fresh start somewhere else.

In the meantime, we saw Cristina’s prior traumas revisit her at the same time she was attempting to support Owen through his recovery process. Cristina found herself unexpectedly drawn to the child of a female cardio patient. As she movingly talked to the child about her mother’s illness Cristina seemed to find her own peace around her father’s tragic death in a car accident when she was nine years old. Strikingly, Cristina chose to stay with the child while her mother was rushed into surgery, a decision that seemed to show Cristina’s increased comfort with balancing the emotional side of her life with her professional goals.

Cristina’s attempt to reassure the child was eerily reminiscent of Owen’s earlier conversation with the cancer patient’s husband, further illustrating how both Owen and Cristina faced parallel journeys. Cristina told the child, ‘First you’ll feel you could have done more to help her but it’s not true. You did everything you could. It won’t feel that way but remember me telling you this, you did everything you could…(and) over time it will hurt less and less.’ Facing this watershed moment in her emotional growth, Cristina shattered and could only find comfort with Owen. In a touching scene, Owen held Cristina lovingly in his arms as she sobbed for her own childhood loss and managed to finally tell him, ‘I miss my dad.’

Building on the promise of this quiet moment of connection, while attending a party at Meredith and Derek’s house Owen asked Cristina a pivotal question. ‘Let’s move in together…I ran it by my shrink and she thinks it’s a good idea. You, me, get our own place?’ Cristina’s heartfelt agreement made it clear she too was ready for the future. The next morning Meredith couldn’t keep herself from advising Cristina that before taking this next step she should be sure there wasn’t more than meets the eye to Owen’s friendship with Teddy. After initially ignoring her friend’s words Cristina spent the day working with Owen and Teddy, observing them through the lens of Meredith’s murmurings and a pair of patients and their story of long lost love regained now that circumstances brought them both to the hospital for treatment.

Further trying to protect her friend, Meredith also urged Owen to talk to Cristina about his attempt to sway Derek’s decision about the cardio position. In the ensuing conversation in a hospital stairwell Owen’s uncertainty of how to incorporate both his memories of the war and his bond with Teddy into his present life in Seattle crashed into Cristina’s own scrambled thinking. As Owen admitted that Teddy both triggered and confused him, paired as she was in his mind with memories of injured soldiers, Cristina grew fearful and withdrew. Added to the tension level was the realization that Teddy had been standing on the stairs below them as Owen and Cristina exposed their innermost fears to each other.

Headed into the season finale the question of how to proceed became further mired in the past for both Owen and Cristina. Each had exposed long-protected wounds and had, in the process, uncovered baggage they had done their level best to ignore was still with them. And once these intense and painful issues had been brought to the surface, distance grew between them at the precise time they had thought they were prepared to step into a deeper shared future. As Owen and Cristina’s fears reached a crescendo so did life at the hospital as into their midst came a grieving widower carrying a gun who sought vengeance for the loss of his wife.

The rapidly dwindling separation of Before and After played out for Owen and Cristina over an increasingly tense landscape. Teddy became offended Owen had tried to influence Derek’s decision about her hospital contract. And given Owen’s disbelieving response to her anger, it seemed their friendship was more tenuous than ever. Soon thereafter, Owen encountered an overtly emotional Cristina who found herself needing validation of the security of their relationship. As each woman asked Owen, in her own way, to make his feelings clear his frustration became palpable. Yet one could see via his tender and anguished responses to Cristina and the brusque way he dismissed Teddy’s concerns that Owen’s frustration was primarily with himself and his inability to step out of his wartime memories in order to fully inhabit his future.

As the gunman attempted to find those responsible for his wife’s death amidst the hospital staff, the situation quickly grew catastrophic. Several of their colleagues were wounded, some were killed. The hospital went into lockdown mode, throwing the orderly system of the facility into chaos. The reality of the situation heightened quickly as Derek was seriously injured while trying to negotiate with the gunman. Saving his life fell to Cristina when it became apparent no attending surgeons would get to them in time as police had cordoned off the building and shut down the elevators. While telling Meredith, ‘for a resident, I’m very good,’ Cristina prepared for the surgery of her life.

Meanwhile, Owen and Teddy who had been in surgery as the lockdown occurred now faced transporting their critical patient to safety outside the hospital. During a tense trip through eerily quiet hospital hallways Teddy made a final attempt to reach Owen. Angry at both himself and the situation his PTSD-related confusion had caused, Owen withdrew himself from the equation and told Teddy he chose no one. Yet once outside the hospital Owen’s feelings became clear even to Teddy as he visually searched the scene while quizzing a police officer on whether medical personnel had been evacuated. Teddy took in Owen’s obvious distress at not being able to ascertain Cristina’s whereabouts. Facing the truth, Teddy turned to Owen and encouraged him to find Cristina, quietly saying, ‘Go…it’s okay, it’s okay to choose.’ Owen nodded his thanks to Teddy for her understanding before running back inside the hospital.

He soon located Cristina in an operating room struggling to save Derek’s life while the gunman threatened her. Owen gathered himself at this most crucial juncture in their lives, entered the OR and set about drawing the gunman’s attention. In his attempt to both secure Cristina time to finish operating and make sure she was safe, Owen stated his feelings plainly and clearly when he said, ‘that is the woman that I love. You touch her, you shoot her, and I will kill you.’ Cristina found her own words after Owen was shot trying to distract the gunman. After the impact of the shot sent Owen to the floor and rendered him temporarily unconscious Cristina quickly advised her friend Meredith to take Owen across the hall to clean and stitch his wound, ‘I’m trying to save your guy now please go try and save mine.’

Later, as Derek’s surgery neared its end we see Meredith and Owen in the operating room alongside Cristina. As Derek’s heart rebounded and beat again in a healthy rhythm Cristina turned and gazed at Owen, exhaustion and intensity plain on both their faces. It’s clear these two have fought their way to the surface and chosen each other in that operating room and I, for one, can hardly wait to see what comes next.

The season premier is entitled “With You I’m Born Again”. The staff tries to deal with physical and emotional trauma in the wake of the vengeful gunman’s deadly rampage; Derek makes a spontaneous decision to resign as chief and return to surgery.

By the way, you can watch The Vampire Diaries Season 2 Episode 3.

Weave a “Freemium” Business Model Element Into Your Consumer Product Marketing Strategy

by: Geoff Ficke

Weave a “Freemium” Business Model Element Into Your Consumer Product Marketing Strategy

A 21st century term applied to a decades old Marketing Concept, Freemium has become one of the most popular vehicles for Consumer Product entrepreneurs, Software designers and Marketers to utilize for increasing sales and distribution of their products. The concept in its most modern form is a spin on the Gift-with-Purchase promotion designed by Estee Lauder Cosmetics in the 1960′s and King Gillette with this safety razor and blade refill strategy first used in the late 19th century.

The word “freemium” is a portmanteau, combining the two aspects of the Business Model, free and premium. The idea is to offer a hook, a free good or service to the consumer, with the Sales Strategy being to up-sell the recipient of the free offer to a premium product or service. This concept is powerful, simple to execute and provides excellent results.

The most compelling word in Consumer Product Marketing is “free”. Who doesn’t want to receive something for nothing? Watch retail store traffic when samples or tastings are being provided. Customers flock to the in-store demonstrator providing the free samples. Politicians make their careers by providing seemingly free benefits to constituents.

The word “premium” is evocative of a better lifestyle, luxury product or enhanced version of a staple product. Most people aspire to add luxury to their environment. A bigger flat screen television, a finer French Perfume, imported rare Extra Virgin Olive Oil or Italian-made shoes are basic examples of the types of products that consumers trend toward as incomes expand. People drive a small Kia automobile for transportation. They drive a Jaguar because they want the premium Features and Benefits that Jaguar Marketing details in its Advertising.

Recently we had a client who was attempting to Market a web-service to the portion of the population who were nearing retirement. Our Product Development team collaborated with software designers to create a free “Club Feature” to entice visitors to the web-site to join instantly. We waived the annual $95 Membership charge. Once aboard, Members were offered a menu of Premium Services, also at Club Member Discounts. The result was an amortization of costs against a broad field of income generators. The site was profitable almost from the date it went on-line.

The Estee Lauder inspired Gift-with-Purchase transformed the Department Store luxury Cosmetic industry. Competitors quickly jumped on this creative Sales Promotion bandwagon. However, Estee Lauder enjoyed the most important element in Consumer Product Marketing and Sales; a first mover advantage. The GWP concept launched the Brand into an orbit that has never been lost.

The Gift-with-Purchase (GWP) Promotional Strategy was followed by the Purchase-with-Purchase (PWP). The GWP offers a free sampler of an assortment of complimentary products. The premium is the minimum purchase required to obtain the GWP. If you do not believe that this technique works, just stand in a Department Store Cosmetic department anywhere in the world when a Beauty Product of Fragrance vendor is offering their GWP.

AOL used a “Freemium” program for years to launch and expand their dial-up internet service. Displays offering free software containing AOL set-up data were ubiquitous in stores and supermarkets. The “freemium” was typically a 90 day no cost teaser trial with a fixed monthly service rate kicking in automatically after the gratis period ended. This model worked magnificently for AOL for a number of years and made the Company the “alpha” service provider that most internet customers initially utilized.

Variants of a “freemium” Marketing Strategy are constantly being utilized. Wellness products, Weight Loss Products, Cosmetics, Food and Drink Products, and a wide range of Consumer Services employ some form of a “freemium” in their Brand building Marketing Campaigns. Infomercials often offer a free trail size of the product with the client paying only shipping and handling. Of course, these offers are almost always accompanied with a Negative Option Feature that kicks on automatically unless you remember to opt out.

Free offers work, but in order to provide goods and services there must be an income stream generated by the give-away. Smart Consumer Product Marketers and Services Providers have learned to craft Marketing Programs that include a free offer with a premium up-sell link. Freemium’s have been around for decades, only the word is new.

The Perks Of Colored Contact Lenses

Colored contact lenses can be a fun way to accessorize your outfit. Colored contact lenses can even be worn by people who don’t need to have their vision corrected. They come in a large number of colors. Contacts come in a visible tint which makes finding them if dropped a lot easier. Also handling them is a lot easier.

Dropping a clear contact can be a nightmare to find but with tinted contacts your life will be a lot more enjoyable. Contacts with their huge selection of colors make it a lot easier to change your eye color slowly. A slight color difference can make wearing contacts more enjoyable. There are a lot of people who are unaware of the large color assortment available for contact lenses and the fact that just about any prescription can be used for these contacts.

A lot of people are still hesitant to try colored contact lenses because they don’t want to shock the people they know by changing their eye color. If you want to wear colored contacts but don’t need your vision corrected you will still need a prescription from an optician or ophthalmologist.

These prescriptions are called plan prescriptions, they will measure your eye width and curvature to make sure the contact fits right. If you share your contacts you will be leaving yourself and the other person open to bacteria that can cause a lot of problems. Since your contact lenses are measured to fit your eye they could cause irritation for someone else to wear.

To drastically change the color of your eyes you need what is called an opaque contacts these are actually tinted to add color to your eyes. Enhancement contacts add tint to your eyes without changing the color. For people who spend a lot of time outdoors you may want to get the UV protection tint contacts.

They help reduce the glare on sunny days. The transitional contact lenses are a great development in contact lenses and gets darker and lighter due to whether you’re indoors or outdoors. Eyes are an important part of anyone’s look so get colored lenses to really dress up your look.

Hitachi Laptop

Fixing the problem of laptop not start

If your laptop does not start at all or it starts but will not boot properly, it is possible that you can fix the laptop at home without taking it to a repair center. Here are some troubleshooting tips for you. I am not sure if I can cover all situation at once, so I will update this post as more examples come into my mind.

Situation 1.

The laptop appears to be dead. You plug the AC adapter but the LEDs (power light, hard drive light, battery charge light, etc.) do not light up and the laptop will not react at all if you press on the power button.

First of all in this situation check the AC adapter. You can test the output voltage with a voltmeter. If you cannot do that, find a known good AC adapter and use it for testing the laptop. It is possible the laptop appears to be dead because the AC adapter is bad (and the battery is discharged). If you know that the AC adapter is working properly and it outputs correct voltage but the laptop is still dead, most likely you have a power issue on the motherboard (or power board on some laptops) and it has to be replaced.
If you have to replace the AC adapter, make sure you use a correct one. The output voltage must be exactly the same as on the original adapter. The output amperage has to be the same as on the original adapter or higher, but not lower.

Situation 2.

When you plug the AC adapter the power LED and the battery charge LED light up. When you press on the power button the laptop powers up but will not start. There is no video on LCD or external monitor.

If the power LED lights up it indicates that the laptop is getting power from the AC adapter. Most likely there is nothing wrong with the adapter but just in case test it with a voltmeter to make sure the output voltage is correct.
Also try this. Unplug the AC adapter, remove the battery such as dell 1691P battery, dell 75UYF battery and dell G5345 battery and wait for 1-2 minutes. After that plug the AC adapter ans try starting the laptop again. Sometimes this trick helps.
It also could be a memory related problem. Try reseating the memory module, just remove it from the slot on the motherboard and install it back. Try installing the memory module into the other slot (if it is available). If you have two memory modules installed, try removing them one by one and start the laptop just with one memory module installed.
If the laptop starts fine with one memory module in both slots, but will not start with the second memory module in both slots, the second memory module is faulty. Replace the module.
If the laptop starts fine with both memory modules when they are installed in the slot A, but will not start with both memory modules installed in the slot B, the slot B is faulty and you will have to replace the motherboard or use only one memory slot.

Situation 3.

When you press on the power button, the laptop makes a series or short and long beeps and will not start up. There is no video on the screen.

In this situation test the memory module as I described in the situation2. Try installing a known good memory module. Most likely you are getting a beep error because of a faulty memory.

Situation 4.

You start the laptop. It sounds like the laptop is booting normally (hard drive LED is flashing) but there is no video on the screen

In this situation test the laptop with an external monitor. If the external screen works fine but there is no video on the laptop LCD, most likely there is a problem inside the laptop display assembly. Go to my previous post witch covers laptop video problems in more details.

Situation 5.

You start the laptop and it starts making repetitive clicking noise or grinding noise.

Most likely you hear this noise because of a faulty hard drive. You can remove the hard drive and start the laptop without it. If the noise is gone, the hard drive is your problem. Replace it.
If the laptop makes clicking or grinding noises and you still have video on the screen, you can run a hard drive test utility. I usually use Hitachi’s drive fitness test. This test is reliable and easy to use.

Situation 6.

The laptop boots into Windows ans works for a while, but after that it shuts down by itself without any reason or warning. You restart the laptop but the same problem appears again

Most likely it is a heat related issue. Listen for the cooling fan, make sure it works.
Also this problem might appear because of a faulty memory module, try some tips from the situation 2.
The laptop still boots ans you still can see the screen, so you can run the memory test. I usually use Memtest86+. Run the memory test and if it fails, replace the faulty module.

Situation 6

The laptop starts normally but video on the screen has lines, some strange characters or other defects

It could indicate a problem with the laptop LCD screen, video cable, graphics card or motherboard. Here’s my previous post witch covers troubleshooting bad images on the screen in more details.

Epidermal Growth Factor Receptor

Function

EGFR (epidermal growth factor receptor) exists on the cell surface and is activated by binding of its specific ligands, including epidermal growth factor and transforming growth factor (TGF) (note, a full list of the ligands able to activate EGFR and other members of the ErbB family is given in the ErbB article). ErbB2 has no known direct activating ligand, and may be in an activated state constitutively or become active upon heterodimerization with other family members such as EGFR.

Upon activation by its growth factor ligands, EGFR undergoes a transition from an inactive monomeric form to an active homodimer – although there is some evidence that preformed inactive dimers may also exist before ligand binding[citation needed]. In addition to forming homodimers after ligand binding, EGFR may pair with another member of the ErbB receptor family, such as ErbB2/Her2/neu, to create an activated heterodimer. There is also evidence to suggest that clusters of activated EGFRs form, although it remains unclear whether this clustering is important for activation itself or occurs subsequent to activation of individual dimers[citation needed].

Diagram of the EGF receptor highlighting important domains

EGFR dimerization stimulates its intrinsic intracellular protein-tyrosine kinase activity. As a result, autophosphorylation of several tyrosine (Y) residues in the C-terminal domain of EGFR occurs. These include Y992, Y1045, Y1068, Y1148 and Y1173 as shown in the diagram to the left. This autophosphorylation elicits downstream activation and signaling by several other proteins that associate with the phosphorylated tyrosines through their own phosphotyrosine-binding SH2 domains. These downstream signaling proteins initiate several signal transduction cascades, principally the MAPK, Akt and JNK pathways, leading to DNA synthesis and cell proliferation. Such proteins modulate phenotypes such as cell migration, adhesion, and proliferation. Activation of the receptor is important for the innate immune response in human skin . The kinase domain of EGFR can also cross-phosphorylate tyrosine residues of other receptors it is aggregated with, and can itself be activated in that manner.

Clinical applications

Mutations that lead to EGFR over[removed]known as upregulation) or overactivity have been associated with a number of cancers, including lung cancer, anal cancers and glioblastoma multiforme. In this latter case a more or less specific mutation of EGFR, called EGFRvIII is often observed. Mutations, amplifications or misregulations of EGFR or family members are implicated in about 30% of all epithelial cancers.

Mutations involving EGFR could lead to its constant activation which could result in uncontrolled cell division a predisposition for cancer. Consequently, mutations of EGFR have been identified in several types of cancer, and it is the target of an expanding class of anticancer therapies.

The identification of EGFR as an oncogene has led to the development of anticancer therapeutics directed against EGFR, including gefitinib and erlotinib for lung cancer, and cetuximab for colon cancer.

Many therapeutic approaches are aimed at the EGFR. Cetuximab and panitumumab are examples of monoclonal antibody inhibitors. However the former is of the IgG1 type, the latter of the IgG2 type; consequences on antibody dependent cellular cytotoxicity can be quite different. Other monoclonals in clinical development are zalutumumab, nimotuzumab, and matuzumab. Gefitinib, erlotinib, and lapatinib (mixed EGFR and ERBB2 inhibitor) are examples of small molecule kinase inhibitors. The monoclonal antibodies block the extracellular ligand binding domain. With the binding site blocked, signal molecules can no longer attach there and activate the tyrosine kinase. Another method is using small molecules to inhibit the EGFR tyrosine kinase, which is on the cytoplasmic side of the receptor. Without kinase activity, EGFR is unable to activate itself, which is a prerequisite for binding of downstream adaptor proteins. Ostensibly by halting the signaling cascade in cells that rely on this pathway for growth, tumor proliferation and migration is diminished. There are several quantitative methods available that use protein phosphorylation detection to identify EGFR family inhibitors.

Efficient conversion of strongly absorbed light by plasmonic gold nanoparticles to heat energy and their easy bioconjugation suggest their use as selective photothermal agents in molecular cancer cell targeting. Two oral squamous carcinoma cell lines (HSC 313 and HOC 3 Clone 8) and one benign epithelial cell line (HaCaT) were incubated with anti-epithelial growth factor receptor (EGFR) antibody conjugated gold nanoparticles and then exposed to continuous visible argon ion laser at 514 nm. It is found that the malignant cells require less than half the laser energy to be killed than the benign cells after incubation with anti-EGFR antibody conjugated Au nanoparticles. No photothermal destruction is observed for all types of cells in the absence of nanoparticles at four times energy required to kill the malignant cells with anti-EGFR/Au conjugates bonded. Au nanoparticles thus offer a novel class of selective photothermal agents using a CW laser at low powers.

In July 2007 it was discovered that the blood clotting protein fibrinogen activates EGFR, thereby blocking regrowth of injured neuronal cells in the spine. Other natural inhibitors include potato carboxypeptidase inhibitor (PCI), which contains a small cysteine-rich module, called a T-knot scaffold, that is shared by several different protein families, including the EGF family. Structural similarities with these factors can explain the antagonistic effect of PCI.

EGFR and Lung Cancer

New drugs such as Tarceva directly target the EGFR. Patients have been divided into EGFR positive and negative, based upon whether a tissue test shows a mutation. EGFR positive patients have shown an impressive 60% response rate which exceeds the response rate for conventional chemotherapy.[citation needed]

Interactions

Epidermal growth factor receptor has been shown to interact with PLCG1, NCK1, Janus kinase 2, CDC25A, MUC1, Caveolin 1, STAT5A, PTPN1, CRK, SHC1, Beta-catenin, PTPN11, PTPN6, STAT1, CBLC, Src, Androgen receptor, STAT3, GRB14, Grb2, PLSCR1, Wiskott-Aldrich syndrome protein, SH2D3A, Epidermal growth factor, CBLB, Cbl gene, ARF4, PKC alpha, SOS1, SH3KBP1, Caveolin 3, Decorin, NCK2 and Ubiquitin C.
